Is there a way to limit access to a project, so the list can be updated without any testers getting into the project during that time without deactivating the project or doing a lot of tester access changes for this?

1) If I remove a user from ALM, who has been creating test cases, req, defects etc how does ALM handle that? does it error out if I click on their assigned test cases or defects? 2) Instead of removing a user, if I make the user inactive - how does ALM handle that Vs removing?

Some QC/LAM camps suggest that you should just deactivate unused users not delete them. I suppose this is in case you need them again later?
