Open your Automail settings in Project Customization (Tools -> Customize-> Automail). Based on your description, you should see 'Assigned To' and 'Status' fields selected under ''Mail On Change Of:'' in your Fields tab.

Now select the Condition tab. Make sure ''All Defects'' is enabled for both your ''Detected By'' and ''Assigned To'' user groups. This will trigger the Automail to send to the user who created the defect as well as the user who was assigned to the defect.
